Cannot connect DB in SQL Server 2005

I uploaded Bank.mdf in database folder
I am using the following string in web.config

<appSettings>

<add key=ConString value=Data Source=.\SQLExpress;Persist Security Info=True;Integrated Security=SSPI;Initial Catalog=pritam6012_Bank;/>

</appSettings>

Am i doing something wrong??

iam getting the following error..
Cannot open database pritam6012_Bank requested by the login. The login failed. Login failed for user 'DOTNETSPIDER1\pritam6012'.
