Can you use User Instance=True?

Can you use User Instance=True? I asked a question about a connectivity issue, but didn't get many replies. So I am curious if I should have this code in the web.config connection and try to work the error out or if I should take it out and try something else. For any questions about my error please refer to my post Not sure if an IIS issue or something web.config for my DB
